I would first inspect the tire to see if it is starting to have a separated tread or any bubbles in tread or sidewall of tire.

Also look at tires to see if have any dry rot cracks indicating tire may have to b changed. If not sure about tire just jack up that tire and spin and look at tire.
Yes fix a flat will cause issue for sure. The liquid will cause imbalance and make noise.
You will need to have tire removed to get out fix a flat and either have tire patched or replaced if worn out or damaged.
